Being an inland boater, Nellie May spends some time on the trailer, and for compliance with local anti-invasive species laws, the hull plug gets put in and taken out frequently. I use Teflon tape on the threads...I guess kinda to prevent wear.

We are embarking on the Great Loop in a week or so, and the hull plug will be "in" for a year or so. Sould I use teflon tape, anti-seize or something else (or nothing) on the threads?

You will get different opinions. I was advised to use Teflon tape by a RT dealer and have been doing so for the past few years. No problem removing the plug after 6 months in the water. Just make it good and snug, overtightening can lead to them being extremely hard to remove. If you use anti-seize it should be rated for marine use.
 
Probably gonna go with the teflon tape. Marine-rated anti-seize is pretty much impossible to get here in South Dakota a million miles from the ocean, and my procrastination kinda prevents a mail order...
